中文摘要: 我国糖尿病发病率居世界首位,已成为重大的公共卫生问题。文献报道称,低等生物沉默信息调节因子(Sir2)的人类同系物Sirt6的全身敲除小鼠会出现低血糖、皮下脂肪减少等表现。而肝脏特异性敲除Sirt6小鼠则表现为脂肪肝,可见Sirt6对糖脂代谢有着非常重要的作用。近年来,GLP-1类似物成为糖尿病治疗领域的新热点,能改善肝脏糖脂代谢。在我们课题组的前面一项国家自然科学基金项目中,也进一步证实了GLP-1受体在肝细胞中的表达。本研究拟通过:采用肝脏Sirt6基因抑制小鼠模型及沉默/过表达Sirt6基因肝细胞模型,通过基因过表达和沉默等分子生物学技术。从动物、细胞及分子水平明确Sirt6在GLP-1改善肝脏糖代谢过程中的作用,为药物的临床应用提供新的理论依据。

英文摘要: The incidence of diabetes in our country is higher than anywhere in the world, it has become a major public health problem. As reports, the human sirtuin 6(Sirt6) konck-out mice displayed persistent hypoglycemia and subcutaneous fat loss. The liver-specific Sirt6 knockout mice manifested as fatty liver, so it played an important role on glucose and lipid metabolism. In recent years, GLP-1 analogs become the new hot spot in the field of treating diabetes, affecting metabolism in the liver. In our previous study, we also confirmed the expression of GLP-1 recptor in liver cells. We thus plan to structure liver specific Sirt6 knockdown mouse model and knockdown/overexpression of Sirt6 gene hepatocyte model, using molecular biology techniques including gene overexpression/silencing, to investigate the mechanism of Sirt6 to interact with the effect of GLP-1 on glycometabolism in liver at individual, cellular and molecular level, and to provide a new idea for the clinical application of GLP-1.
