中文摘要: Wnt通路转录因子b-catenin蛋白是维持干细胞全能性的关键分子,传统观点认为TCFs利用b-catenin的转录活性起始了多种干细胞特异性基因的表达。然而最新研究表明b-catenin的转录活性并不参与干细胞全能性的维持,因此TCFs如何起始基因转录成为干细胞研究中的一个令人感兴趣的问题。申请人发现一个新的细胞核蛋白p44BL,能够不依赖b-catenin的存在与TCFs协同促进基因转录; 同时,p44BL表达于干细胞中,并显著促进干细胞特异性因子Nanog、Oct-4和Sox-2的表达。这表明p44BL是潜在的TCFs家族蛋白的转录共激活子和干细胞全能性的调控者。因此申请人希望通过本项目的研究,揭示TCFs-p44BL这一全新转录激活模式在Wnt信号通路和干细胞自我更新中的调控作用,为进一步利用p44bl基因进行诱导多功能干细胞的研究提供分子基础。

英文摘要: b-catenin is the downstream effector of Wnt signaling pathway, which plays a crucial role in stem cells renewal. Classically, TCFs family transcriptional factors employ the transcriptional domain of b-catenin to initiate transcription of a variety of stem cell harbored genes. However, recent studies suggested that b-catenin utilizes a transcription-independent manner to maintain the pluripotency of stem cell. Now, it is coming to be an interesting question how TCFs regulate gene transcription in stem cells. In the preliminary studies, we discovered a novel nuclear protein p44BL, which harbors gene transcriptional activity and stimulates gene transcription together with TCFs in a b-catenin-independent manner. Meanwhile, p44BL is expressed in stem cells and greatly increases the transcription of Nanog, Oct-4 and Sox-2. Thus p44BL might be a co-activator of TCFs family transcriptional factors and a potential regulator of stem cell pluripotency. In this project, we hope to reveal the regulation mechanism and biological function of TCFs-p44BL in the Wnt signaling pathway and stem cell renewal, and we also hope these findings could benefit iPS studies in the future.
