中文摘要: 真姬菇是一种极具开发价值的珍稀食用真菌,有着独特的营养价值和抗肿瘤、提高免疫力等功效。多糖是食用菌中最具免疫增强潜力的成分,然而目前关于多糖免疫活性的研究正面临着调控机理及构效关系等亟待解决的瓶颈问题。本课题对真姬菇多糖进行分离纯化和结构鉴定,研究单糖组成、糖苷键连接方式、分子量及空间构象等分子结构对其免疫调控活性的影响。同时以小鼠单核巨噬细胞Raw264.7为细胞模型,从免疫学和生物学角度研究多糖对巨噬细胞中细胞信息传递分子NO及肿瘤坏子因子α(TNF-α)、白细胞介素-1β (IL-1β) 等重要细胞因子的影响,并采用先进的分子生物学手段,从转录水平上研究多糖对各细胞因子mRNA表达的影响,进一步从基因表达水平上揭示真姬菇多糖免疫调控机制。 本研究有望从分子水平上对真姬菇多糖免疫调控机制及构效关系提出新的观点,为合理开发和充分利用真姬菇资源奠定扎实可靠的理论基础和科学依据。

英文摘要: Hypsizigus marmoreus (Peck) Bigelow is one kind of rare mushroom which have been valued as edible and medicinal resources for its abundant nutrition and various biological activities such as anticancer and immunomodulatory activity. Polysaccharides are the best known and most potent mushroom-derived substances with immunomodulating properties. However, the immumomodulatory mechanism and the structure-activity relationship of polysaccharides are still not well established. This study investigate the effect of molecular structures of polysaccharides such as monosaccharides composition, glycosidic linkage, molecular weigh on the immunomodulatory activity. Furthermore, the mechanism of polysaccharides from H. marmoreus on mice macrophages was discussed by investigating from the point of NO and cytokines (TNF-α, IL-1β, et al.) secretion. We further investigate the mechanism of immunomodulatory activity of polysaccharides at the transcriptive level through the gene expressions of these cytokine by RT-PCR. This study will propose a new viewpoint on the mechanism of immunomodulatory activity and structure-activity relationship of polysacchrides from H. marmoreus laying the reliable theoretical foundation and scientific basis for H. marmoreus resource rational-exploitation.
