中文摘要: 赤霉病是小麦的一种主要病害,对其产量和品质均有影响。虽然已经定位多个抗赤霉病QTL,但未克隆到抗赤霉病基因。当前用于抗赤霉病转基因研究的多是一些防卫基因和抗菌肽,这些基因的转基因植株在大田条件下抗性并不理想。因此,克隆新的抗赤霉病关键基因,阐释抗赤霉病机制就显得尤为重要。Jacalin 相关凝集素因响应生物/非生物胁迫以及SA 和JA 等防卫信号分子刺激的特性受到研究者的广泛关注。TaJRL2是我们从抗赤霉病种质望水白中克隆的一个凝集素类基因,该基因主要在穗部的颖壳和外稃中表达。在抗病种质中受赤霉菌诱导上调表达,在感病材料中没有因赤霉菌诱导而上调。VIGS 技术沉默抗病种质中TaJRL2 后,其抗赤霉病能明显下降。转TaJRL2 超量表达载体的小麦对赤霉菌抗性得到提高。本项目拟从遗传、分子和表型水平进一步明确TaJRL2 参与赤霉病抗性反应的功能,丰富赤霉病抗性改良的理论基础和技术途径。

英文摘要: Fusarium head blight (FHB) or scab is a kind of devastating wheat disease worldwide, which result in great yield losses and poor grain quality. Several QTLs for FHB resistance have been located, but no resistant gene has been identified.Now, most of genes transformed to wheat to improve the FHB resistance are kinds of defense genes or antibacterial peptides. The transgenic wheat using these genes haven't showed ideal resistance to FHB. So that, identification of new anti-disease gene and clarification its mechanism showed more important. Jacalin-related-lectin (JRL) genes have drawn wide attention due to its response to abiotic or biotic stimuli and to defense signal molecular. A jacalin-related-lectin gene (TaJRL2) that is mainly expressed in glume and lemma was identified from spike during flowering stage in Wangshuibai, wheat germplasm resistant to FHB. The expression of TaJRL2 was up-regulated significantly in resistant variety Wangshuibai after inoculated by fusarium grminearum, while the phenomenon didn't appear in susceptible variety Mianyang87-19. Silencing of TaJRL2 in Wangshuibai using virus-induced gene silencing (VIGS), and resistance of FHB reduced remarkably.
