中文摘要: 原花青素是苹果多酚中主要活性物质,但其在苹果果实中的合成机理尚不完全清楚。本项目拟在前期发现的苹果MdLAR1和MdANR2基因表达水平与果实原花青素含量显著相关的基础上,继续发掘这两个基因的等位变异,同时构建苹果资源果实原花青素含量基础数据库,采用候选基因关联分析方法鉴定核心变异序列,开发MdLAR1和MdANR2的功能基因标签。此外,通过基因酶活或启动子活性等分析验证核心变异序列对基因功能或表达水平的影响,阐明MdLAR1和MdANR2不同等位基因及其互作调控苹果果实原花青素积累的分子遗传基础。本项目的开展既能深入了解影响苹果果实原花青素积累的分子机理,又可获得控制果实原花青素含量的功能分子标记用于苹果果实品质遗传改良,具有重要的理论意义和应用价值。

英文摘要: Proanthocyanidins (PAs) are the major bioactive component of apple polyphenol, however, the mechanism underlying the biosyntehsis of PAs in apple fruit is unclear. More recently, we have found that the expression level of MdLAR1 and MdANR2 correlates significantly with the PA content. In this project, we will further explore the allelic variation of these two genes. A fruit PA content database of apple germplasm will be constructed for candidate gene-based association mapping to identify the core allelic variation(s), which will be used to develop functional molecular markers of MdLAR1 and MdANR2. In addition, the effect of the core allelic variation in promoter or coding region on gene functionality will be validated by measurement of its corresponding promoter activity or enzyme activity, respectively. It will be clarified that the genetic contribution of the alleles of MdLAR1 and MdANR2 and their combination to PA accumulation in apple fruit. .This project possesses a theoretical significance in expanding our knowledge of molecular mechanism of PA accumulation in apple fruit, and will also provide functional molecular markers useful for genetic improvement of apple fruit quality.
