中文摘要: 生物如何适应不同的环境是进化生物学一直探索的重要问题之一。部分陆生被子植物在进化过程中受到了水生环境的选择压力,最终形成了水生被子植物。毛茛属水生类群起源于该属陆生类群。本项目拟采用毛茛属水生类群水毛茛 (Ranunculus bungei)、北京水毛茛 (R. pekinense),沼生类群石龙芮 (R. sceleratus),及陆生类群鸟足毛茛 (R. brotherusii)、茴茴蒜 (R. cantoniensis) 的转录组数据,开展以下工作:(1) 筛选出水毛茛、北京水毛茛、石龙芮在适应水生/沼生环境过程中受正选择压力的基因,并分析其功能和代谢通路;(2) 选择毛茛属水生类群受正选择压力的部分基因,克隆基因全长并转化拟南芥,进行水淹胁迫试验以验证基因功能。本项目对理解植物适应水生环境的分子机制,发掘具有提高植物水淹耐受力的基因具有重要意义。

英文摘要: Understanding how organisms have adapted to different environments is a long-standing goal of evolutionary biology. Some terrestrial angiosperms have experienced selective pressures from the aquatic habitat, and evolved to aquatic angiosperms. Aquatic Ranunculus is originated from the terrestrial Ranunculus relatives. In this study, we plan to use transcriptome data of two aquatic plant Ranunculus bungei, R. pekinense, one marsh plant R. sceleratus, and two terrestrial plant R. brotherusii and R. cantoniensis. Our objects are to (1) identify the positively selected genes, which might be involved in adaptation to aquatic habitats, from R. bungei, R. brotherusii and R. cantoniensis; (2) determine whether the genes can improve waterlogging tolerance of Arabidopsis thaliana by overexpressing them in the species. This study will help us to understand the molecular mechanism of adaptation to aquatic habitats, and explore genetic resources for improving plant tolerance to waterlogging.
