中文摘要: 内源性一氧化碳分子在生物体内起着信号传导和生理调节功能。荧光探针的制备对于监测这一活性分子的生理分布以及研究其作用机制具有重要科学意义和研究价值。本项目将一氧化碳与环钯配合物的配位反应特性与荧光分析相结合,拟合成几种水溶性的荧光功能化环钯配合物,研究其与一氧化碳的羰基化反应性能,探索将化学反应过程转变为荧光信号输出的原理和途径。(1)构筑设计一类能够与一氧化碳快速计量反应的模型环钯配合物,探索配合物的结构与性能的关系;(2)将模型钯配合物的反应位点与荧光基团结合制备有机荧光配体,利用碳氢键活化反应将有机配体与钯反应得到水溶性环钯配合物荧光探针并对其结构进行表征;(3)通过荧光开关机制,建立一氧化碳剂量与荧光信号强度之间的对应关系,实现低浓度一氧化碳的快速荧光信号响应和可视化成像。本项目的完成将为合成荧光功能化环钯配合物提供思路,为针对内源性一氧化碳荧光探针的设计奠定理论和实验基础。

英文摘要: As an important gasotransmitter molecule, CO is proposed to play significant roles in modulating responses to both chemical and physical stresses.The development of sensitive CO-responsive fluorescent probe would meet a critical need for new technologies which has great importance and application value. The carbonylation of palladacycle with CO has been investigated deeply and was used for organic synthesis and catalytic chemistry with high yield and wild reaction requirement. We aim to design and synthesize kinds of water soluble fluorescent functionalized palladacycles, anticipating that the rapid carbonylation reaction between palladacycle and CO would correspond to the change of fluorescent signal. By screening rapid CO-responsive model palladacycles and combining the reaction site with water soluble fluorophore, the fluorescent ligands were obtained. Then the ligands react with Pd(Ⅱ) complexes using typical organometallic reaction such as C-H activation to give target water soluble palladacycle probe with low fluorescent background which would be characterized by NMR and X-ray crystal diffraction. Through the construction of fluorescent switch, it will establish the relationship between CO dose and fluorescence intensity to achieve the rapid detection for CO at low concentration.
