中文摘要: 长链非编码RNA(lncRNA)是近年发现的对基因表达起调控作用的内源性核酸分子,在细胞增殖、分化和凋亡等过程中起重要作用,lncRNA异常表达与肿瘤有关。本项目拟通过lncRNA表达谱及靶标信息分析阐释纳米氧化锌颗粒(ZnO NPs)选择性诱导肿瘤细胞毒性效应的分子机制。本研究中,利用lncRNA芯片及定量PCR检测lncRNA和mRNA的特征性差异表达,并应用双向聚类分析等方法构建lncRNA和mRNA共表达谱,进行ZnO NPs选择靶向肿瘤细胞毒效应有关的信号调控研究。应用生物信息学和数学计算工具建立lncRNA的基因调控网络模型,探索lncRNA在选择细胞毒性中的作用,预测lncRNA靶标分子。本项目将揭示lncRNA在ZnO NPs靶向杀伤肿瘤细胞中的作用和信号途径,为阐明ZnO NPs的细胞毒效应机制提供新资料,也可为肿瘤治疗提供新的靶点信息。

英文摘要: Long non-coding RNAs (lncRNAs) are emerging as new endogenous nucleic acid molecules to regulate genes expressions. LncRNAs play important roles in the cell proliferation, differentiation and apoptosis. The abnormal expressions of lncRNAs are associated with tumor. To explore the molecular mechanisms of selective cytotoxicity between tumor cells and normal cells induced by zinc oxide nanoparticles (ZnO NPs), we analyze lncRNAs expression profiles and their targets information. In the study, we investigate the specific differential expression of both lncRNAs and mRNA by the lncRNA microarry and the quantitative PCR. To study the signaling regulations of tumor cell-targeted cytotoxicity, we establish lncRNAs expressions profiles using bicluster analysis and other methods, and build the lncRNAs genetic regulatory networks model to explore the role of lncRNA in the selective cytotoxicity, forecasting target molecules of lncRNAs by bioinformatics and algorithm analysis. The project will reveal the roles and the signaling pathways of lncRNAs in the targeted killing of tumor cells by ZnO NPs, and provide new information to clarify the cytotoxicity mechanisms of ZnO NPs, and also provide new targets for cancer treatment.
