中文摘要: 牛体内脂肪沉积与脂肪细胞的增殖分化和葡萄糖摄取密切相关。然而,目前关于牛脂肪沉积相关分子机理尚不明确。本项目拟建立由牛成熟脂肪细胞去分化得到的高度均一的前体脂肪细胞(DFAT)为模型,从新的角度,更精细地研究并揭示牛脂肪形成机制。基于DFAT模型,比较皮下和肌内脂肪来源的牛DFAT细胞增殖分化能力差异并找出相关基因时序表达规律;通过腺病毒载体上调、下调NCOR1基因表达,从mRNA、蛋白水平分析牛DFAT细胞分化相关基因变化规律和葡萄糖摄取量变化,并进一步通过免疫共沉淀、蛋白磷酸化检测以及蛋白酶抑制剂阻断实验等方法探究NCOR1是否通过PPARγ和胰岛素相关通路作用,并初步阐明相应的作用机制。上述研究结果将为进一步阐明牛脂肪形成机制奠定一定基础,并为今后培育低体脂、高肌内脂肪肉牛功能基因育种提供一定的理论依据。

英文摘要: The fat deposition of bovine is closely related to the adipose cell proliferation, differentiation and glucose uptake. However, the mechanism of bovine fat deposition regulation is undefined. The expected goal of our research is to establish uniform cell model (DFAT) via dedifferentiating mature fat cells, from a new perspective, which more refined research and reflect regulation of adipogenesis. Based on the DFAT model, analyze the proliferation and differentiation differences of preadipocytes from different parts of cattle body and to explore the corresponding molecular mechanisms. Meanwhile, we are also planning to reveal the discipline of bovine preadipocytes proliferation,differentiation and the glucose uptake regulated by NCOR1, at mRNA level and protein level. Future more, find out its potential signal pathways via phosphorylation detection,Co-Immunoprecipitation and inhibitor blockage experiments. The final purpose of our project is laid the foundation of to clarify the mechanism of bovine adipogenesis. Otherwise, provide theoretical basis to breeding new beef cattle breeds of low subcutaneous fat but high intramuscular fat.
