中文摘要: APOBEC3s家族不仅有广泛的抗病毒作用,而且参与了多种肿瘤恶性生物学行为的调控。最近文献报道APOBEC3s家族可以调控HPV病毒基因的转录和表达,可能通过诱导HPV基因特定区域的高突变,参与HPV相关的宫颈癌的发生。高危型HPV持续感染是宫颈癌发生的必要因素,本课题利用TR-PCR和Western blot 技术分析APOBEC3s家族各成员在维吾尔族妇女宫颈各级别病变的基因和蛋白水平表达,通过3D-PCR技术检测APOBEC3s高表达宫颈病变组织中HPV16 DNA LCR的突变情况,在体内验证APOBEC3s对HPV DNA的编辑作用,并检测病变组织中HPV的整合状态,分析APOBEC3s与宫颈病变病情进展的相关性,在HPV感染转归、整合中的作用,将丰富对APOBEC3s在HPV抗病毒、参与宫颈肿瘤细胞调控的认识;为HPV感染患者随访、宫颈癌诊治提供依据。

英文摘要: APOBEC3s not only have a wide range of antiviral activity, but is also involved in the regulation of a variety of malignant biological behavior. Recent literatures had reported that APOBEC3s family can regulate gene transcription and expression of the HPV virus, possibly by inducing the high HPV gene mutations of specific area involved in the occurrence of HPV-related cervical cancer. Continuing high-risk HPV infection is a necessary factor for cervical cancer. Using TR-PCR and Western blot technical analyses APOBEC3s family gene and protein expressing levels in Uygur women with cervical lesions; using 3D-PCR technology analyses mutant case in HPV16 DNA LCR, E2 region at APOBEC3s in vitro expression.detect mutations in HPV16 DNA LCR, E2 region in cervical lesions of APOBEC3s high expression By 3D-PCR technology, to confirm the role of APOBEC3s editing for HPV DNA in vivo, and to detect the integration state of HPV in the lesions and the expression of P53, analyze correlation of APOBEC3s and progression of cervical lesions and the role of APOBEC3s in the outcomes of HPV infection, the integration of the HPV. It enriches the knowledge of APOBEC3s about the role of antiviral HPV and the participation of Regulatory functions of cervical cancer cells; provide the basis for follow-up and treatment. of patients with HPV infection.
