中文摘要: 蛋白激酶GSK3β和核受体RXRα在体内发挥着重要的生物学功能和病理学作用,但是它们之间的相互调节以及这种调节在肿瘤中的作用至今无人报道。我们最近发现GSK3β能够介导RXRα的磷酸化,并且能够调节RXRα的N端缺失体tRXRα的产生。这揭示GSK3β和RXRα之间存在相互作用,而这种相互作用可能会产生重要的生物学功能。本项目将在分子、细胞和动物水平上,利用多学科交叉的实验方法,研究GSK3β对RXRα的磷酸化调节作用,以及这种磷酸化调节对RXRα的生物学功能的影响。进一步我们还将研究GSK3β介导的RXRα磷酸化在肿瘤发生和发展过程中的作用。本课题的开展将揭示一个新的RXRα的活性调节方式和机制,发现一个新的GSK3β的功能展现方式,加深我们对RXRα的基因型和非基因型活性的认知。同时本课题还将增加我们对RXRα和GSK3β在肿瘤发展中功能的理解,为肿瘤的治疗提供理论依据。

英文摘要: Glycogen Synthase Kinase 3 beta (GSK3β) and Retinoid X Receptor alpha (RXRα) play important and complicated roles in the biological and pathological processes, especially in tumor initiation and progression. However, their reciprocal regulation and the function of the regulation in cells and during tumor development are not known. Our prelimilary results indicate that GSK3β phosphorylates RXRα and also induces the production of tRXRα, an N-terminal deleted RXRα, indicating the interaction of GSK3β and RXRα. The objective of this proposal is to identify the regulation and the regulatory mode of RXRα phosphorylation mediated by GSK3β and the role of the phosphorylation in tumor development. In this proposed studies, we will apply integrated multidisciplinary approaches to focus on studying these questions, 1) How does GSK3β induce the phosphorylation of RXRα? 2) How does phosphorylation regulate the genomic and nongenomic function of RXRα? 3) Do RXRα and tRXRα mediate the effect of GSK3β during tumor development? 4) Does GSK3β mediate the crosstalk between some signal pathways and RXRα? Our proposed studies will reveal a new regulatory mode of RXRα function and an unidentified substrate of GSK3β.
