中文摘要: 基于通路水平的遗传影像策略探寻遗忘型轻度认知障碍(Amnestic mild cognitive impairment, aMCI)的神经网络损害特征已成为神经科学领域的研究热点,尤其是tau蛋白代谢通路基因多态性与多模态fMRI技术融合为切入点揭示aMCI 转化为阿尔茨海默病(Alzheimer's disese,AD)的神经影像病理机制在国内外研究尚处于起步阶段。因此,本研究团队将基于在aMCI研究领域的前期积累,结合多维度认知功能评估及多模态fMRI,在中国汉族人群中同期选择aMCI及其匹配健康对照组人群,分析tau蛋白代谢通路基因多态性及其集聚效应,建立基于tau蛋白代谢通路水平的aMCI神经网络损害模式,寻找遗传风险因素影响aMCI向AD转化的影像学表征模型。

英文摘要: The characteristics of altered neural networks in amnestic mild cognitive impairment (aMCI) patients with a risk factor for Alzheimer's disease (AD) based on genetic image strategy which has become the hotspot in the field of neuroscience. Especially, little is known about neuroimging pathological mechanisms of aMCI through the association between genetic polymorphisms of tau protein pathways and multi-modality fMRI. Therefore, in the present project, we recruited the aMCI patients and matched controls with age, gender and education. Multi-dimension neuropsychological tests and multi-modality fMRI were used to explore the cognitive function and nerural networks in these subjects. Then, genetic polymorphisms of tau protein pathways and combined effect of these multi-genetic polymorphisms were evaluated. The project was designed to reveal the altered patterns of neural networks based on the level of tau protein pathways. Moreover, the imaging characterization model of the progression of aMCI to AD will be explored.
