中文摘要: 由高温诱导产生的血红素加氧酶1(HO-1)又称热休克蛋白32,是一种具有抗凋亡和细胞保护作用的抗氧化防御酶,但其对高温诱导的牛卵泡颗粒细胞凋亡的作用机制尚不清楚。基于研究组前期发现HO-1是通过p38信号通路对大鼠卵巢发挥保护作用,故本研究拟首先通过在体实验研究高温及非高温环境牛卵巢HO-1基因及其下游p38信号通路表达的变化,进而通过离体实验体外培养牛卵泡颗粒细胞,并通过激活或阻抑HO-1基因研究高温应激状态下卵泡颗粒细胞HO-1基因及其下游p38信号通路表达变化,同时进一步分析HO-1代谢产物信号分子CO对p38信号通路及雌激素分泌的调节,旨在揭示HO-1对高温诱导的牛卵巢颗粒细胞凋亡的作用机制,为高温应激环境下奶牛颗粒细胞凋亡的诱因和机制提供理论支撑。

英文摘要: Hemeoxygenase 1 (HO-1/HSP32) play an anti-apoptotic and cytoprotective effects in various cells, but little is known about its definitive function in granulosa cells of cows. We have found the protective role of HO-1 for ovary of rat via p38 signaling pathway. To clarify the underlying role of HO-1 in granulosa cells of cows , HO-1 gene expression and related signals involved in p38 signaling pathway in ovary and culture granulosa cells were analysed; expression of HO-1 was silenced or activated in the granulosa cells using small interfering RNAs (siRNAs) or HO-1 inducer to further verify the functional role of HO-1 through p38 signaling pathway mediated in granulosa cells; meanwhile, the levels of gas signaling molecule and E2 in cultured media were determined to further clarify the mechanism of HO-1 on granulosa cells of cows via p38 signaling pathway. The research seeks to provide a theoretical basis for possible causes and regulation mechanism of granulosa cells apoptosis in summer environment.
