中文摘要: 退行性病变是中老年群体肌骨系统最常见的疾病,尤其多发于膝、髋关节及腰椎。深入系统地研究退行性病变异常骨结构的结构形态和力学性能,及其功能适应性过程,对于弄清退行性病变的发病机理,以及寻找相应的预防和治疗措施有重要意义,是非常重要的应用基础研究课题。本项目以胫骨、股骨和腰椎三个部位发生退行性病变的异常骨结构为研究对象,通过多层次的计算生物力学方法(非线性显微有限元分析、骨功能适应性过程模拟预测)、计算机影像技术(Micro-CT扫描、扫描电镜、原子力显微镜技术)和力学实验方法(宏观非线性力学实验、纳米压痕实验)相结合,研究各种异常骨结构内部细观层次和微纳观层次的显微结构形态和力学性能;定量模拟预测各种异常骨结构的功能适应性过程,探索各种异常骨结构的形成机理;最后寻找各种异常骨结构多层次力学性能与其结构形态的关系。为弄清退行性病变的发病机理,以及寻找相应的预防和治疗措施做出贡献。

英文摘要: Degenerative diseases are very common for the musculoskeletal system of the elderly population, especially at knee joint, hip joint and lumbar spine. It is of great significance to deeply investigate the morphology, mechanical property, as well as the functional adaptation process of abnormal bone structure caused by degenerative bone disease for a better clarification of its pathology, and prevention & treatment strategy. It is a very important applied basic research topic. The abnormal degenerative tibia, femur and lumbar vertebral body are the subjects in this project. By integrating multi-level computational biomechanical methods (i.e. nonlinear micro-finite element analysis and simulation on bone functional adaptation process), imaging techniques (Micro-CT scanning, scanning electronic microscopy, and atomic force microscopy techniques) and mechanical testing methods (macro-nonlinear mechanical test and nanoindentation test), the internal microstructures and mechanical properties of each abnormal structure at meso-, micro-, and nano-levels will be investigated, the functional adaptation process of each abnormal bone structure will be quantitatively predicted to explore the formation mechanism of each abnormal bone structure. Finally, for each abnormal bone structure the relationships between the multi-level mechanical properties and its morphologies will be investigated. This project may contribute to elucidate the pathology of degenerative bone diseases and find the corresponding preventive and curative strategies.
