中文摘要: 肿瘤细胞及其相关生物分子的检测,在肿瘤的预警和诊断及提高肿瘤患者生存率方面具有及其重要的作用。但是生物样品中,许多肿瘤细胞及其相关生物分子的浓度极低,因此对它们的测定需要有极高的灵敏度和选择性。本课题拟开展酶循环放大信号提高石英晶体微天平分析灵敏度的工作,并将其用于肿瘤细胞及其相关生物分子检测的应用研究。主要研究工作包括如下几方面:(1)构建纳米生物探针提高芯片系统灵敏度;(2)滚环复制放大策略检测肿瘤细胞表面生物活性分子;(3)工具酶循环放大检测肿瘤细胞及细胞内活性分子。为生物学及临床医学等领域提供重要的参考依据,也为肿瘤的早期诊断提供新方法和新技术。

英文摘要: The detection of cancer cell and its related biomolecules plays an essential role in the precaution、diagnosis and the survival rate of important diseases such as cancer. However, in biological samples, the concentrations of cancer cell and its related biomolecules are normally very low. As a result, the detection of cancer cell and its related biomolecules need to be highly sensitive and selective. This study intends to carry out the research of quartz crystal microbalance analysis with enzyme cycle signal amplification and its application in cancer cell and its related biomolecules detection. The main research can be divided in three aspects: (1) the design of nano bioprobe and improvement of chip system; (2) rolling copy amplification for the detection of cancer cell surface biomolecules; (3) tool enzyme cycle amplification for the detection of cancer cell and intracellular biomolecules. The propoesd study may offer new approaches for senstive and selective biomolecule analysis for early cancer diagnosis.
