中文摘要: 前期研究中,将核酸适体技术与光致电化学手段相结合,构建新型光致电化学传感器。研究了光电转换界面的制备及多种传感器的组装方法,取得了较丰富的理论成果。目前,发表影响因子大于5.0的SCI论文4篇,申请发明专利两项。 下一步工作计划从两方面展开:一是利用新材料组装高效光电传感界面,进一步提高传感器的灵敏度;二是将这种灵敏方法应用于疾病的早期诊断中。本申请基于这两个目的,采用新型材料和传感器的组装方法,将高灵敏的光电技术应用于与糖类抗原相关的肿瘤标志物的高灵敏检测中。 研究表明,大多数癌变细胞的表面都会发现糖蛋白中糖基的改变。本课题设计利用与肿瘤相关的糖抗原检测抗体。首先制备高效光电转换界面,以常见的单糖为模型,建立光致电化学检测方法。然后采用该方法研究几种复杂寡糖抗原对抗体的特异性识别,并将这种高灵敏、高特异性的方法应用于患者血清的检测中,实现与糖类抗原相关的肿瘤标志物的高灵检测。

英文摘要: In previous research work, we integrated the aptamer with photoelectrochemistry establishing some novel photoelectrochemical biosensors. The preparation of photoelectric interface and photoelectrochemical biosensor were investigated and relatively rich theoretical results were obtained. At present, published 4 SCI articles impact factor greater than 5.0, and applied for two invention patents. The next step of work will be carried out from two aspects. On the one hand, new type of material will be used to construct photoelectric interface of high efficiency. On the other hand, this sensitive method will be applied to the early diagnosis of disease. This application was designed based on these aims. Since the change of carbohydrate group in glycoprotein occurs in most cancer cells. We will apply the highly sensitive photoelectrochemical methods to investigate the interaction between carbohydrate and protein. The main goal is to realize the detection of tumor related antibody using carbohydrate antigen. Firstly, commonly used monosaccharide and lectin are selected as object of study, establishing photoelectrochemical detection methods.
