中文摘要: 脂肪组织内源性芳香化酶产生的雌激素是男性及绝经后女性雌激素的主要来源。我们在前期发现perilipin 基因敲除小鼠(脂肪营养不良)脂肪内芳香化酶表达显著下降的基础上,拟观察机体不同部位包括附睾、宫旁、腹股沟、大网膜、肾周、乳腺及棕色脂肪组织中芳香化酶的细胞定位、蛋白表达及活性改变;进而研究过表达或敲低芳香化酶对脂肪分化及分解产生影响的可能机制,以及脂肪分化过程中芳香化酶表达及活性的变化;以肥胖和脂肪营养不良小鼠为动物模型,应用芳香化酶抑制剂或腺病毒过表达芳香化酶,观察脂肪含量、脂肪分化、脂肪分解的改变以及血脂异常和胰岛素敏感性的变化。通过以上研究,探讨脂肪组织芳香化酶在不同部位表达及活性的不同介导了局部脂肪组织雌激素水平的不同,及糖皮质激素、TNF-α 等刺激局部雌激素产生的反应不同,进而导致局部脂肪组织分化及分解的失平衡,引起脂肪组织在全身的重新分布并导致全身代谢紊乱的分子机制。

英文摘要: The major source of endogenous estrogen in men or in postmenopausal wowen is from adipose tissue. Our previous work has found that aromatase expression was significantly reduced in adipose tissue in perilipin knockout mice ( a lipodystrophy mouse model). Based on our findings, we plan to investigate aromatase expression, activity and cell localization in different adipose tissues, including the epidigymic, parametrial, inguinal, omental, perirenal, breast and brown adipose tissue. The effect of aromatase on pre-adipose cell differentiation will be investigated when we overexpress or knockdown the expression level of aromatase in the cells. And aromatase expression and activity will also be detected at different stages during the differentiation of pre-adipose cells. Overexpress or suppress aromatase expression in adipocyte, to to investigate the possible role of aromatase in lipolysis while treatment with glucocorticoid, TNF-α, isoproterenol and insulin. Overexpress or suppress aromatase expression in db/db obesity mice or perilipin knockout mice, to check the changes of fat mass, dyslipidemia and insulin resistance.
