中文摘要: 神经介素S(NMS)是一种含有36个氨基酸并与受体结合发挥多种生理功能的活性肽。研究表明NMS和受体在免疫细胞均有分布,而其对猪的免疫的调节途径和作用如何未见报道。本研究采用流式细胞术和格里斯法(Gris)等方法以探索NMS对培养的猪肺泡巨噬细胞和脾淋巴细胞的功能及对猪肺泡巨噬细胞和淋巴细胞上NMS受体的mRNA和蛋白表达的影响为切入点,探讨其参与猪免疫调节的途径,进一步研究静脉注射NMS对猪小肠粘膜上皮的M细胞的数量分布和NMS受体的基因和蛋白水平的影响,以研究其在抗原转化、加工和呈递过程中的作用,深入探讨静脉注射NMS对猪免疫器官和血清中细胞因子和NMS受体的表达及分泌的影响。总之,上述的研究内容旨在验证NMS是否作为一种重要的免疫信息传递物质,参与猪体内的免疫调节作用。

英文摘要: Neuromedin S (NMS) was a 36 amino acid peptide, which can play many physiological functions by combinding with its receptors. All the reported researches have demonstrated that the distribution of NMS and its receptor in the immune cell, but the information about the regulation of the immune system remained unclear. Base the effect of NMS on the immune function of cultured pig alveolar macrophages and lymphocytes, this project set to explore the passable way which NMS may participate in the immune regulation in pig; further explain the expression of mRNA and protein of NMS receptors and the numbers and distribution of M cell of intestinal epithelial under intravenous injection with NMS to investigate its effect in the process of the transformation, antigen processing and presentation;mainly explore the expression level of gene and protein of NMS receptors and cytokines in immune organs and the secretion of cytokines in serum by intravenous injection with NMS to pigs using the flow cytometry and Gris methods etc. In brief, the goal of all the mentioned above is to verify whether NMS can act as an important immune transmitter to be involved in the immunoregulatory sysytem in pigs.
