中文摘要: 盐胁迫是造成农作物减产的非生物胁迫之一。改良和提高作物耐盐性是开发利用盐渍化土地、保证粮食产量稳定增长的重要措施。黍稷是一种优良的耐盐作物,种质中蕴藏着丰富的耐盐基因资源有待挖掘利用。本课题组通过室内芽、苗期鉴定与田间全生育期鉴定相结合的方法从黍稷核心种质中筛选出39份综合耐盐性状优良的材料,并对其进行了生理生化水平的耐盐机理研究。本项目在此基础之上,拟选用两份耐盐性差异显著的材料,在二叶一心期利用300mM混合盐溶液进行0h、0.5h和24h的胁迫处理,采用高通量测序技术分析其在响应盐胁迫过程中的基因表达情况,构建转录组水平耐盐调控网络,初步揭示黍稷耐盐分子机理;挖掘黍稷耐盐转录因子并进行基因表达和转基因功能验证。这一工作的开展,将首次从分子水平探讨黍稷耐盐机理,并有望挖掘出与黍稷耐盐性密切相关的转录因子基因资源,为后续黍稷耐盐工作的深入研究提供理论和物质保障。

英文摘要: Salinity is an important abiotic stress that adversely affect plant growth and development and has a crucial impact on agricultural productivity and yield. Thus, it urgently requires the breeding of crops with increased salt tolerance and the discovery of salt-adaptive genes. Broomcorn millet (Panicum miliaceum L.) is a salt-tolerance species and has higher salt tolerance than most of crops, but little is known about the molecular basis of salt tolerance. Our laboratory focused on the identification of salt tolerance varieties of broomcorn millet by the study of germination and seedling stage in laboratory and reproductive stages in field and have found some tolerance varieties. In this study, RNA-seq method will be used to explore the transcriptome change of broomcorn millet under 300 mM NaCl+Na2SO4 stress solution maintained 0h, 0.5h and 24h respectively to initially reveal the molecular mechanism of sal tolerance. Furthermore, we will isolate and clone salt tolerance-related of transcription factor genes and analysis the gene function and expression pattern.
