中文摘要: 金属硫蛋白(Metallothionein,MT)是一类低分子量、富含半胱氨酸的小分子蛋白。由于其分子结构的特异性,金属硫蛋白可特异、稳定、高效地吸附多种重金属(Cd2+、Hg2+、Pb2+等)。微藻细胞具有适应性强、生长迅速、对重金属耐受性强等特点,是一种新颖的重金属废水生物修复材料。 本项目将利用一年左右的时间,围绕转金属硫蛋白基因微藻净化重金属废水的几个关键科学问题开展研究: (1)利用不同的表达质粒、启动子和 MT 基因,选用不同的转化方法和微藻细胞种类,构建转 MT基因工程微藻; (2)利用 PCR 和 Western Blotting 等技术,鉴别转 MT 基因微藻,测定金属硫蛋白表达量; (3)通过测定其对不同重金属离子的耐受性与吸附性能,筛选得到转金属硫蛋白基因工程微 藻。 课题的研究成果将为重金属废水的净化与生态修复开辟新的思路,并为转 MT 基因工程微藻的产业化应用打下基础,具有重要的学术和应用价值。

英文摘要: Metallothionein is a kind of cysteine-rich protein with low molecular weight. Because of its specific molecular structure, metallothionein can adsorbe various kinds of heavy metals such as Cd2, Hg2, Pb2 specifically and efficiently. Microalgae is a class of novel biological remediation material for heavy metals wastewater because it has the characteristics of strong adaptability, rapid growth and strong tolerance for heavy metals. The project will spend a year or so to carry out research on the following key scientific problems centering on the purification of heavy metal waste water by transgenetic microalgae with metallothionein gene: (1) Construction of transgenetic microalgae with metallothionein gene using different transformation methods, expression plasmid, promoter, metallothionein gene, transformation methods and micrlalgal species; (2) Identification of transgenetic microalgae and determination of expression levels of metallothionein gene through PCR and Western Blotting techniques; (3) Screening of microalgae strains with metallothionein gene by determinating its tolerance of different heavy metal ions and its adsorption capability. The project's results will not only open up new ideas for heavy metals wastewater purification and ecological restoration, but also lay the foundation for industrial applications of microalgae with metallothionein gene, so it has important academic and practical value.
