中文摘要: IL-17R/ACT1信号通路是新近发现的一条在抵抗病原感染的免疫防御和炎症反应过程中发挥重要作用的免疫信号通路,在脊椎动物中普遍存在且高度保守。然而,在海洋无脊椎动物中是否存在这种信号通路一直是未知之谜。幸运的是,我们在太平洋牡蛎中发现了IL-17R/ACT1的部分序列,证明了在软体动物中存在IL-17R/ACT1的同源基因。但是,太平洋牡蛎中IL-17R/ACT1是否具有类似脊椎动物的功能和信号通路仍然不得而知,这也构成了本申请的研究内容。本研究拟克隆出其全长序列,分析其在抵御病原侵袭过程中的时空表达模式;利用RNAi和免疫共沉淀等技术解析IL-17R/ACT1信号通路,探讨它们在牡蛎天然免疫中的功能。因此,本研究对于阐明IL-17R/ACT1信号通路在海洋无脊椎动物天然免疫的功能和分子机制具有重要的价值,而且对于理解和探索IL-17R/ACT1信号通路的起源和演化规律有着深远的意义。

英文摘要: IL-17R/ACT1 signaling pathway is a recently discovered immune signaling pathway, which plays a key role in the process of resistance to exogenous pathogens infection defense and inflammatory response. They are ubiquitous and highly conserved among vertebrates. However, the evidence of this signaling pathway in marine invertebrates remains mysterious. To our surprise, a partial sequences of IL-17R/ACT1 gene were obtained from the Pacific oyster genome and EST databases, indicating the existence of IL-17R/ACT1 of homologous genes in marine mollusks. Nevertheless, whether IL-17R/ACT1 gene in the Pacific oyster share similar functions or signaling pathway is still unknown, which constitutes the main contents of this study. In this study, we plan to study expression pattern of Pacific oyster IL-17R/ACT1 gene during different pathogenic microorganisms and PAMPs stimulation, clarifying their roles in resisting invasion of pathogens. Meanwhile, to explore their innate immune function in the Pacific oyster, we intend to use Pull-down and coprecipitation methods to analyze IL-17R/ACT1 signal transduction pathways. Therefore, our results will not only contribute to understanding the molecular mechanism of IL-17R/ACT1 signaling pathway in innate immunity of marine invertebrate, but also provide a new insight into its origin and evolutionary rule.
