中文摘要: 本课题以中医药理论为指导,针对在自然基金资助下经过系统药代动力学研究的药典经方冠心丹参方,引入机器学习与数据挖掘领域的先进理论阐释方证相应配伍机制的科学内涵。首先对气滞血瘀型胸痹证动物进行多证型方剂治疗,并采集功效指标样本,构造层次小波概要数据结构对多模态、异构、异分布样本进行归一化表征,建立功效指标稀疏条件随机场模型,并依据网络拓扑关系划分气滞血瘀型胸痹证的功效指标集。进而根据疾病进程(进展期、急性期、恢复期)研究冠心丹参方组方变化(药味配伍变化、药量配比变化)状态下的功效指标集的关联性变化,并依据同步检测的成分群药代轮廓特征,实现基于病程-组方-功效指标集-药代轮廓特征的动态稀疏条件随机场建模,实现从病-证-方-药的动态体系共同揭示冠心丹参方组方的深层配伍机制,为诠释传统中医药方证相应配伍理论的丰富内涵提供有力的科学数据支持,为精细化预测合理治疗方案作出有益的尝试。

英文摘要: Under the traditional Chinese medicine (TCM) theories, Guanxin Danshen Decoction (GDD), the classic TCM prescription, has been with systematic pharmacokinetic studies granted by the National Natural Science Foundation in the prophase work stage. Now, it will be further studied on the correspondence of prescription and syndrome in the current project by the theory and method of machine learning and data mining. The current project intends to put the focus on the systematic and scientific explanation for TCM compatibility in GDD, via researching on the TCM effect profiles of active ingredient group of GDD, evaluating the weight factor for progression of disease, then, developing the new mathematics models, ie. Conditional Random Field (CRF) with the data features of multimodal, heterogeneous and different distribution, to reveal the qi stagnancy and blood stasis characteristics of correspondence of prescription and syndrome of GDD. By exploring the metabolism characteristics and the TCM action of the effective components, we try to clarify the substance foundation of prescription action in vivo. Via studying the mathematical model of topological relation of CRF, and the relationship of disease - syndrome - prescription - TCM, we intend to elucidate the reasonable and scientific TCM compatibility theories in the aspects of correspondence of prescription and syndrome of GDD, and aim at achieving the purpose of providing new modes for the TCM prescription study and convincing scientific data to support the rich connotation of the TCM compatibility theories.
