中文摘要: 本课题依据非线性复杂适应系统科学原理及研究思路,借鉴药物动力学、药效动力学结合模型(PK/PD结合模型),以黄连解毒汤为实验体系,研究"药效作用"与"化学组成"的动态变化规律,采用SUM算法、最小二乘法等数据挖掘技术,构建黄连解毒汤主要指标性成分与抗血管性痴呆火热证之间的数学方程;借鉴整合药动学的研究方法,建立黄连解毒汤复方分类整合药动学与药效学之间的相关性分析,以期在中医药整体观理论指导下,阐明中医理论"泻火解毒"与"内毒蕴积、热伤脑络"的"方证相应"关系,探索建立复方组合预测与筛选数学模型,从而准确表达"证"模型证候群影响"方"中指标性成分体内动力学过程,准确预报"方"中指标性成分的作用程度及作用靶点,为指导临床用药提供理论依据和技术手段,为中药复方体系实现"表征参数检测- - 针对适应证进行复方组合预测与筛选- - 优化临床方案"提供一种模拟模式。

英文摘要: Based on the scientific principles and research ideas of a non-linear complex adaptive system, this research was aimed to study the dynamic change laws between the active effects and the chemical compositions on the experimental system of Huanglianjiedu decoction. By means of pharmacokenetics- pharmacodynamic (PK-PD) modeling and data mining, this research was intended to establish the mathematical model predicting and screening the active combinations developing from Traditional Chinese Medicine compound. Meanwhile, the integrated pharmacokinetic methods was applied to estsblish a correlation analysis between the classified constituents derived from Huanglianjiedu decoction and pharmacological effects. These research methods were used to indicate exactly how the syndrome groups in the "syndrome" influence the pharmacodynamic process of indicative components in the "TCM compound", thereby predict accurately the effect degrees and effect targets of indicative components in the "TCM compound". Therefore, the mathematical model can be applied as theoretical basis and technological methods for guiding clinical treatment. In this research, the PK-PD model and the integrated pharmacokinetic methods were used to probe into the study field of "prescriptions corresponding to syndromes". The aim of this research was to provide a simulated model of "characteristic parameter detecting - predicting and screening the active combinations from TCM compound according to different syndromes - providing excellent clinical treatment" on the complex system of TCM compound.
