中文摘要: 骨缺损是骨科临床上的常见病,如何促进其愈合始终是骨科研究的一大焦点。骨髓间充质干细胞(BMSCs)是骨组织工程首选的种子细胞,其定向迁移及向成骨分化在骨缺损修复中发挥了重要作用。我们通过前期研究发现麝香可以促进BMSCs向骨缺损处迁移,其迁移与促进骨缺损处趋化因子的表达有关。而信号通路P38MAPK、PI-PLC及PKC与BMSCs迁移关系密切,为进一步探讨麝香对于信号通路的调节机制,本项目通过血清药理学方法,体外阻断P38MAPK、PI-PLC及PKC信号通路以观察麝香对高迁移率族蛋白1(HMGB1)、基质细胞衍生因子(SDF-1)、干细胞因子(SCF)的影响,用骨缺损模型观察麝香促进BMSCs迁移的分子机制,从而为中药临床治疗骨缺损提供理论依据。

英文摘要: bone defect are the clinical common diseases in Orthopaedics. How to promote its healing is a great focus to the scholar's study. Bone marrow mesenchymal stem cells (BMSCs) to migrate and ossification in the bone defect to develop an important function in the repaired, we preliminary discovered the musk can promote BMSCs migrate to the bone defect at expect study.BMSCs migrate hand in hand with opening of related signal way, therefore we calculate that the musk can help BMSCs's migration by opening the P38MAPK、PI-PLC and PKC pathway. we will observe the influence of musk to the P38MAPKse、 PI-PLCses and PKC signal pathway by blocking and through serum pharmacological method,and observe the molecular mechanism of the musk helps the BMSCs to migrate by bone defects model.The result will comprehensivelly reveal the migration mechanism. to provide scientific theory to clinic application for bone defects.
