中文摘要: 研究表明:激活AMPK活性,抑制肝脏过度糖异生(gluconeogenesis),减少内源性葡萄糖生成,是治疗2型糖尿病的重要靶标之一。据报道:桔梗皂苷通过激活AMPK活性而能够预防酒精性大鼠脂肪肝和氧化应激损伤。我们研究亦发现:桔梗皂苷具有明显的降血糖作用,可以改善葡萄糖耐量,增加肝糖原含量。因此,我们推测:肝脏是桔梗皂苷发挥生物活性的重要靶器官之一,桔梗皂苷可能通过激活肝脏AMPK活性,抑制肝糖异生信号通路而发挥降血糖作用。 本项目基于大量的前期研究工作及桔梗皂苷的降血糖作用。为进一步阐明桔梗皂苷的降血糖分子调控机理,拟以桔梗皂苷中的代表化合物-桔梗皂苷D(platycodin D, PD)为研究对象,观察其对高脂饮食联合STZ诱导的2型糖尿病小鼠的降血糖作用,结合HepG2细胞糖生成实验,在动物和细胞水平阐释桔梗皂苷D对AMPK/PEPCK/G-6-Pase肝糖异生信号通路中的调控。

英文摘要: AMP-Activated protein kinase (AMPK), an energy sensor that regulates cellular metabolism, has been reported to be involved in the glucose metabolism of various key tissues such as liver, skeletal muscle, adipose tissues, and pancreatic β cells in the pathogenesis of T2DM. Previous findings showed that AMPK plays a key role in the control of hepatic metabolism. Therefore, AMPK could control blood glucose levels by inhibiting hepatic glucose production. Our results indicated hypoglycemic effect of crude saponins from Platycodon grandiflorm on type 2 diabetes mice. We hypothesized that platycodins exhibited hypoglycemic activity via activation of AMPK and suppression of hepatic gluconeogenesis. In this work, based on the above findings, platycodin D (a major triterpenoid-type saponins in P. grandiflorm) was employed to investigate hypoglycemic effect of type 2 diabetes mice in order to explore the regulation mechanism on AMPK/PEPCK/G-6-Pase signal passageway of hepatic gluconeogenesis. These expect results will supply theory basis for utilizing platycodins and P. grandiflorum as new supplementary treatment agency for type 2 diabetes.
