中文摘要: 氧化应激导致白癜风发生,但是机制并不清楚,许多研究发现白癜风患者存在黑素细胞凋亡,线粒体损伤等现象,由此我们提出假设氧化应激可能通过线粒体通路引起黑素细胞凋亡进而导致了白癜风发生。滋补肝肾方以滋补肝肾为立法,是北京中医医院治疗白癜风的有效方剂。本课题以过氧化氢诱导黑素细胞凋亡模型为研究对象,观察该方及拆方的含药血清及主要成分对黑素细胞氧化损伤致黑素细胞凋亡、细胞内钙稳态、线粒体跨膜电位以及凋亡蛋白等的影响;探索氧化应激是否通过线粒体通路引起黑素细胞凋亡,该方是否作用于线粒体信号转导通路来调控氧化损伤引起的黑素细胞凋亡,揭示滋补肝肾治则的作用环节,为临床治疗白癜风提供科学依据。

英文摘要: Oxidative stress cause the vitiligo, but the mechanism is unclear.Many studies have found that patients with vitiligo may have phenomenon such melanocytes apoptosis and mitochondrial damage, so we put forward the hypothesis that oxidative stress could lead to to vitiligo by mitochondrial pathway ..The prescription of nourishing liver and kidney is an effective prescription of vitiligon from Beijing Chinese Medicine Hospital. .This subject is melanocytes of apoptosis induced by hydrogen peroxide ,to explore whether oxidative stress lead to melanocytes apoptosis by mitochondrial pathway, to research mechanisms of the prescription of nourishing liver and kidney from the mitochondrial signal transduction pathway by probing into the influence on apoptosis melanocytes and calcium homeostasis in melanocytes, provide the scientific basis for the clinical treatment of vitiligo.
