中文摘要: 通过联合应用"悬浮培养"与"免疫磁珠分离"相结合的方法对体外培养的前列腺癌细胞进行"癌干细胞特性"(Stemness)富集及分离。成功分离出前列腺癌干细胞并对其进行初步鉴定。利用基因芯片技术筛选出与前列腺癌干细胞特性相关分子信号通路,发现Hedgehog信号通路的激活可以促进前列腺癌干细胞的自我更新能力(Self-renewal capacity)及体内肿瘤起始能力(Tumor-initiating capacity);特异性阻断Hedgehog信号通路可抑制前列腺癌干细胞的体内外"自我更新能力"(Self-renewal capacity)。因此,胚胎前列腺发育和前列腺癌进展过程中重要分子信号通路- - -"Hedgehog信号通路"在前列腺癌干细胞增殖、分化以及肿瘤起始中起着关键性的作用。癌干细胞在前列腺癌发生及恶性进展中的作用及其分子机制的初步研究结果,为阐明前列腺癌发生机制提供新思路,为特异性根治前列腺癌提供新资料,也为探索其它癌症起源及恶性进展的共性提供借鉴,具有重要的理论意义和潜在的临床应用前景。

英文摘要: By using the combination of suspension culture system and immunomagnetic separation to enrich and separate cancer stem cells in culturing prostate cancer cell lines, we have successfully indentified prostate cancer stem cells, which possess the characteristics of self-renewal capacity, chemoresistance and tumor-initiating capacity. Using gene chip technology to screen the potential molecular signaling pathway in the maintain of prostate cancer stem cell properties, we found that the activation of Hedgehog pathway could promote the self-renewal capacity and in vivo tumor-initiating ability of prostate cancer stem cell. Meanwhile, the specific blocking of Hedgehog pathway by GANT61 can significantly inhibit prostate cancer stem cells self-renewal capacity in vivo. Therefore, Hedgehog signaling pathway, the common pathway of the fetal prostate development and prostate cancer progression, play a crucial role in the prostate cancer stem cell proliferation, differentiation and tumorigenesis.
