中文摘要: 研究表明AML的复发和耐药与骨髓基质细胞(BMSC)对AML细胞的保护作用有关,但骨髓基质细胞与白血病细胞相互作用机制目前还不清楚。Hedgehog信号通路是在果蝇中首先发现的,Hedgehog信号通路的异常在恶性肿瘤的形成、生长等方面具有重要的作用。基于以上研究结果,本研究拟从体外细胞、体内动物模型上进行研究,建立体内、外白血病细胞与骨髓基质细胞共培养体系,研究Hedgehog信号通路在骨髓基质细胞对急性髓系白血病细胞凋亡影响中的作用及机制,检测与基质细胞共培养的急性髓系白血病细胞中Hedgehog信号通路中关键基因的表达,及信号通路抑制剂对凋亡相关基因表达的影响。同时建立裸鼠移植瘤模型,检测共培养体系中骨髓基质细胞对白血病细胞的凋亡影响是否通过Hedgehog信号通路起作用,最终通过靶向Hedgehog信号通路抑制逆转BMSC对AML的保护作用,为AML临床治疗提供新的实验依据。

英文摘要: Studies have shown that relapse and drug resistance of AML has the relationship to bone marrow stromal cells (BMSC) for AML cells of protection, but the interaction of bone marrow stromal cells with leukemia cells is still unknown.Hedgehog signaling pathway is first found in fruit flies, Hedgehog signaling pathways plays an important role in the malignant tumor formation、growth and maintain.Based on these researches,this study tries to research from cells in vitro and in vivo animal models, set up the leukemia cells and bone marrow stromal cells cultivation system in vivo and in vitro , study the Hedgehog signaling pathway 's apoptosis effect and action mechanism of bone marrow stromal cells to acute myeloid leukemia cells ,detect the expression of key genes in Hedgehog signaling pathways that the cell in acute myeloid leukemia cells cultured with stromal cells, and the influence of the signaling pathway inhibitor to apoptosis related gene expression. And setting up nude mouse transplantation tumor model, detect whether bone marrow stromal cells influence to apoptosis of leukemia cells by Hedgehog signaling pathways in the co-culture system, ultimately through targeted Hedgehog signaling pathway inhibition and reverse BMSC protection of AML, provide new experimental basis for clinical treatment of AML.
