中文摘要: 本项目以某些重要疾病标志物的特征基因片段(如乳腺癌特征基因序列BRCA1、口腔癌特征基因序列ORAOV1、慢粒细胞白血病(CML)相关基因片段、BCR/ABL融合基因片断、H5N1 病毒相关基因等)为研究对象,基于工具酶技术,开发研制集高灵敏度、高选择性和快速响应功能于一体的电化学生物传感器,为发展适用于食品安全、重大疾病预警与诊断相关靶标分子快速检测的新技术、新方法打下理论与实践基础。本项目主要研究内容包括以下几个方面:(1)基于限制性内切酶技术与纳米探针的DNA电化学生物传感界面的构建;(2)基于切刻内切酶辅助电流放大技术的DNA电化学生物传感界面的构建;(3)基于切刻内切酶及G-四链体电流放大技术的DNA电化学生物传感界面的构建;(4)基于工具酶技术的核酸适配体传感界面的构建。本项目所开发的技术亦可望用于单核苷酸多态性(SNP)、基因分型及药物调控释放的研究。

英文摘要: The aim of this proposal is to develop the highly sensitive, selective and fast response electrochemical biosensors based on using tool enzyme technology for detecting the special gene sequence of the markers of some important diseases, such as the special breast cancer gene sequence BRCA1, DNA species related to oral cancer overexpressed 1 (ORAOV1), Chronic Myelogenous Leukemia (CML) related gene sequence, BCR/ABL fusion gene sequence and H5N1 virus related gene sequence and so on. The proposed new techniques and methods are very useful for food safe, early warning and diagnosis of some important diseases. The main research contents of this project can be conclused as follow: (1) Construction of the DNA electrochemical biosensing interface based on the restriction endonuclease and nano probe technique; (2) Construction of the DNA electrochemical biosensing interface based on the technique of nicking endonuclease assisted current amplification; (3) Construction of the DNA electrochemical biosensing interface based on the nicking endonuclease and G-quadruplex DNAzyme assisted current amplification technique; (4) Construction of the aptamer electrochemical biosensing interface based on the tool enzyme technique.
