中文摘要: 中枢神经系统(CNS)是免疫赦免器官,CNS内引入抗原可引发针对该抗原的负向免疫调控,但关键的启动细胞和效应机制不明确。近年来发现稳态下(Steady state)脑内存在树突状细胞(DC),其生理功能尚不清楚。鉴于DC处于启动和调控获得性免疫反应的中心地位以及具有迁移的特性,我们推测脑内的DC亚群可能参与脑内抗原引发的免疫调控。本项目将"稳态下脑内DC亚群在CNS免疫赦免中的作用"作为核心科学问题,以现代细胞免疫学和分子免疫学实验技术为研究手段,探讨稳态下脑内DC亚群的组成和天然免疫功能状态;以此为基础,进一步探索CNS引入抗原后脑内DC亚群对抗原的摄取和向外周淋巴器官的迁移,以及对获得性免疫反应的调控及相关机制。该研究有助于阐明脑内DC亚群在CNS免疫赦免形成或维护中所扮演的角色,为以后基于DC亚群为靶细胞来修复或重建CNS免疫赦免提供理论上的可能性。

英文摘要: Introduction of antigen into the central nervous system (CNS), the immune privilege organ, can lead to a negative antigen-specific immune response. However, the key cells initiating this response as well as the effector mechanisms during the process remain poorly understood. In recent years, dendritic cells (DCs) have been found located in the brain, but its physiological function is unclear. Taking into account the central role of DCs in triggering and regulating adaptive immune responses as well as their ability to migrate, we hypothesized those DC subsets in the brain may be involved in immune regulation caused by brain antigen. We considered how DC subsets in healthy brain contribute to CNS immune privilege as the core scientific issues. Using modern cellular/molecular immunology experimental techniques, the project aims to investigate the composition of DC subsets in the healthy brain as well as their functional state in innate immunity. On this basis, further study will be performed to investigate the brain DC subsets in antigen uptake, migration into the peripheral lymphoid tissues, and regulation of the adaptive immune response caused by introduction of antigen into CNS.
