中文摘要: 珊瑚微生物学是一个新兴的研究领域,主要研究共附生于珊瑚虫中的微生物与珊瑚虫之间的关系,目的是充分了解珊瑚礁生态系统的特征,并最终防止世界范围内珊瑚礁生态系统的衰退。 本项目从珊瑚共生体的生物固氮功能切入,以三亚湾鹿回头和西沙永兴岛珊瑚礁生态系统为研究对象,珊瑚白化过程中追踪采集珊瑚样品,利用固氮功能基因(nifH)的高通量测序技术,分析珊瑚共附生固氮微生物群落结构变化特征,利用荧光定量PCR (Real-time PCR)研究珊瑚共附生微生物固氮功能基因的表达特征。利用乙炔还原法测定活珊瑚的生物固氮活性,研究活珊瑚固氮效率的变化特点;结合珊瑚周围环境因子的变化,应用多元统计分析方法深入分析珊瑚共附生固氮微生物对珊瑚白化的响应特征,探索珊瑚白化过程中内部高效循环系统的相关信息,研究成果将为进一步研究珊瑚礁生态系统的恢复机制奠定基础,有助于推动我国珊瑚微生物学的发展。

英文摘要: Coral microbiology is a new field, driven largely by a desire to understand the interactions between corals and their associated microorganisms and to use this knowledge to eventually prevent the spread of coral decline. In this project, the response of coral associated nitrogen-fixing microorganism to coral bleaching will be researched in Luhuitou coral reef of Sanya Bay and Yongxing Island coral reef of Xisha Islands. These two coral reef ecosystem were effected by different levels of human activities. Molecular methods such as the massively parallel pyrosequencing technique and quantitative PCR method will be adopted to study the community composition variation and the expression of nifH of coral associated nitrogen-fixing microorganisms. Acetylene reduction method will be applied to detect the nitrogen fixation rates. These rich data sets will provid much detail information about the coral associated nitrogen-fixing microorganism. Multivariate statistical analysis will be performed to explore the relationship bewteen coral associated nitrogen fixing microorganism and environment factors during the processes of coral bleaching.
