中文摘要: 近年来,纳米银的生物毒性已成为纳米毒理领域的研究前沿,其中纳米银的致毒机制和毒性调控两方面研究尤为关键。由于纳米银具有大的比表面积,其生物毒性表现出对表面化学结构强烈的依赖性。因此,本项目拟采用一系列化学结构上系统可比且配位作用明确的简单小分子修饰纳米银,通过调节毒性相关的关键性质(银离子释放和活性氧产生),进而扰动纳米银的生物毒性。进一步比对银离子释放/活性氧产生行为和相关毒性规律的对应关系,得到纳米银的生物毒性机制。同时,总结基于表面化学的纳米银生物毒性的构-效关系,并以此构-效关系为基础,采用具有调控效用结构的多种有机分子修饰纳米银,以调控纳米银的生物毒性行为。本项目的研究结果对于深刻理解纳米银的生物毒性机制及最终实现对纳米银的合理制备和安全使用都具有重要价值。

英文摘要: Recently, the biological toxicity of nanosilver has become the frontier of the nanotoxicological research, especially for the study on mechanism and control of the toxicity. As known, the toxicity of nanosilver shows a strong dependence on the chemical structure of surface, due to the large specific surface area. This project intends to modify the nanosilver with a series of organic molecules, which have a systematical comparability in chemical structure and a defined fashion in interaction with the surface Ag atom. In this way, the toxicity-related properties including Ag+ release and ROS generation are regulated, which would further induce the corresponding changes on the toxicity of nanosilver. Then, by comparing each behavior (Ag+ release or ROS generation) of nanosilver and the toxicity effects, the exact mechanism could be concluded. Meanwhile, the relationship of structure-property on the toxicity of nanosilver could also be acquired by correlating the toxicity effects with the chemical structure of surface. Based on this law, we could control the toxicity of nanosilver by simply modifying the surface with multiple effectively-structured molecules.
