中文摘要: 随着蛋白质组学的发展,对蛋白质翻译后修饰的研究受到人们越来越多的重视。蛋白磷酸化是一种最重要的蛋白翻译后修饰,它调控着诸多基本的细胞过程,比如基因表达、信号传导和蛋白降解等过程。因此磷酸蛋白的检测和磷酸化位点的确定对于开发新的药物靶点和了解疾病机理具有重要意义。由于体系中磷酸蛋白的数量和蛋白磷酸化程度相对较低,一般磷酸蛋白在质谱分析前要先经过富集过程。本项目前期研究发现Chitosan-GMA-IDA-Fe (III)纳米材料对于磷酸蛋白有高特异性的捕获能力。本项目以生物兼容性好的壳聚糖类纳米微球为富集手段,以方便快速的常压敞开式质谱为分析工具,来研究对磷酸多肽的选择性检测和鉴定。本项目期望通过DESI及reactive DESI分析模式的摸索,开发一种快速方便的磷酸多肽的富集检测方法,为理解蛋白磷酸化在重要的生化路径上起到的作用提供新线索。

英文摘要: With the development of proteomics, more and more attention has been paid to the post-translation modification of proteins. Protein phosphorylation is one of the most important mechanisms to regulate cellular processes such as gene expression and membrane transport. Owing to its highly dynamic nature and low stoichiometry, an enrichment method is required for the MS analysis of protein phosphorylations, which decreases complexity of the samples and increases the detection sensitivity for low abundant phosphoproteins. Our previous data indicates that Chitosan-GMA-IDA-Fe (III), a novel multifunctional nanosphere, exhibits highly specific enrichment of phosphopeptides. With the introduction of a series ambient ionization methods, which allow samples to be directly ionized in the open environment and introduced into the mass spectrometer without the need for sample pretreatment, MS analysis becomes amenable to high-throughput analyses in ambient environment. In this project, by taking the advantages of Chitosan-GMA-IDA-Fe (III) and desorption electrospray ionization (DESI), an ambient ionization method, we plan to develop a rapid, simple and reliable analytical method for specific detection of phosphorylated protein and identification of phosphorylation sites.
