中文摘要: 猪2型链球菌病是一种重要的人畜共患病,给世界养猪业带来了巨大的经济损失,也直接威胁人类的健康。筛选鉴定新型保护性抗原一直是猪2型链球菌免疫致病机理研究和防控研究的热点之一。前期研究应用免疫蛋白质组学的方法并辅之于动物实验从猪2型链球菌表面蛋白成功筛选鉴定了与宿主细胞互作的保护性抗原。但其在致病和免疫方面的功能却缺乏进一步的研究。本研究中,我们鉴定了2个保护性抗原和3个体内诱导抗原,并建立了基于Biacore的蛋白质相互作用研究手段。在后续的工作中,我们进一步以保护性抗原HP0197为研究对象开展了其与宿主细胞的相互作用的研究。初步确定HP0197与宿主细胞表面肝素分子相互作用,为进一步揭示HP0197的生物学功能奠定了基础。

英文摘要: Streptococcus suis serotype 2 (SS2) is an emerging zoonotic agent world-wide causing high economical losses, and it is also a threat to human health. Identification of novel protective antigens is one of the focuses to the researches on pathogenesis and prevention of SS2. We have identified the protective proteins which could interact with the host proteins from the surface proteins of SS2 by immunogenic proteomics and animal experimentation in the previous studies. However, the functions of these proteins in pathogenesis and immunity remain unknown. In this work, 2 protective antigens and 3 in vivo induce antigens have been identified. And a Biacore-based approach, which could be used to investigate the protein-protein interactions, has been developed. In the subsequent works, our works focus on the interactions between protective antigen HP0197 and host cells. The preliminary results indicate that HP0197 could interact with surface heparin of host cells, and this research lay a foundation for further functional study of HP0197.
