中文摘要: 线粒体铁蛋白( MtFt)是一种新的定位于线粒体上和细胞铁代谢密切相关的蛋白,其确切功能还不清楚。而胞内铁稳态的紊乱及由此产生的氧化应激反应和神经退行性疾病如PD等的发生密切相关。本研究以MtFt基因敲除小鼠(MtFt-/-)和过表达MtFt的SH-SY5Y细胞为材料,MPTP处理细胞或小鼠制作PD模型,应用免疫组化,酶学,荧光探针标记,western blot,ICP-MS,放射性同位素标记,Real-time PCR,电镜,核染色,流式细胞术,膜片钳等技术,研究MtFt在小鼠不同脑区的表达情况及在PD发生中的功能,目的是在分子水平上深入了解和掌控MtFt在铁代谢、氧化应激,神经退行性疾病及细胞凋亡中的功能和作用,阐明MtFt在调节铁代谢过程中对神经退行性疾病保护作用的分子机制及信号通路。本工作将对今后和铁代谢相关的疾病如PD的预防和治疗提供新的策略,具有重要的理论意义和现实意义。

英文摘要: Mitochondrial ferritin (MtFt) is a new protein which locates in mitochondrial and is related to cellular iron metabolism. The function of MtFt is not clear by now.Disorder of cellular iron homeostasis and oxidative stress are involved in some nerve degeneration such as PD,In this item,we use MtFt-/- mouse and SH-SY5Y cells overexpressed MtFt as materials which will be treated with MPTP to make PD model,then using immunohistochemical method,RNA interference,enzymology,fluorescent probe,western blot ICP-MS radioactive isotope labeling, Real-time PCR, electron microscope, karyotin, flow cytometry and patch clamp to investigate the expression of MtFt in different area of brain and the function of MtFt during PD.The aim of this item is to deeply understand the function and role of MtFt in iron metabolism,oxidative stress,verve degeneration and cell apoptosis, elucidating the protedtive effect of MtFt on iron metabolism and nerve degeneration.This study not only has important significance in thery,but provide a new way and strategy in preventing nerve degeneration related to disorder of iron metabolism in the future.
