中文摘要: 针对生物样品基体复杂、样品量少、待测物含量低等特点,开展生物样品中痕量或超痕量分析物的高选择性、高效的新的样品前处理技术。拟将核酸适配体化学键合有序介孔炭材料,并固载于磁性纳米四氧化三铁上,获得磁分离萃取介质,探索制备新方法,发展通透性好、适合于富水相生物样品高特异性识别,高选择性的适配体萃取介质;表征核酸适配体功能化有序介孔炭材料萃取介质的形貌结构,研究其萃取性能;应用于生物样品中多巴胺、生物素、苯丙氨酸、酪氨酸等组分的分离富集,采用液相色谱、电化学、化学发光等高灵敏检测技术,发展高选择性样品前处理,建立复杂样品的高灵敏快速分析方法。集成核酸适配体的高特异性和有序介孔炭材料高度规整的孔道、孔径可调、大比表面积、易于修饰等特点,发展高特异性、操作简单和快速分离能力的复杂生物样品前处理技术。

英文摘要: It is considered as complex matrix and less sample, and some analytes are found in trace or ultratrace quantities in biological samples. Therefore, it is very significant to develop a selective and effective technology to determine trace analytes in biological sample. The magnetic separation of medium was accepted by the covalent bonding established between the aptamer and the ordered mesoporous carbonaceous materials, it is immobilized on the surface Fe3O4 nanoparticles. Their potential applications were explored to develop new method, good permeability, high specificity and selectivity of aptamer extracting medium in aqueous biological samples, the characterisation of the aptamer functional ordered mesoporous carbonaceous materials was studied, They were applied to separate and enrich for dopamine, biotin, phenylalanine, and tyrosine components in biological samples by chromatography, electrochemical, chemiluminescence, etc. Combine the high specificity of aptamer with highly structured holes, adjustable aperture, large surface and easily modify of ordered mesoporous carbonaceous materials, it is very important to develop high sensitive, speed analysis, and simple operation sample pretreatment technology in complex biological sample.
