中文摘要: 多囊卵巢综合症(PCOS)是严重影响妇女身体健康的重要疾病之一,PCOS患者存在早期动脉粥样硬化(AS),其远期并发症心血管疾病的发生率远远高于正常人群。目前还缺乏有效防治PCOS合并AS安全性较高的药物,且对其发病机制不清楚。我们在前期工作中发现PCOS合并AS模型中主动脉及尿液中氧化应激的标记物明显增加,说明氧化应激参与了此发病机制。原花青素是一种清除人体内自由基最有效的天然、高效的抗氧化剂,本研究采用高脂饮食饲养双氢睾酮(DHT)诱导的PCOS大鼠16周,建立PCOS合并AS模型,利用放射免疫、蛋白质印迹及实时定量PCR等技术,观察原花青素对PCOS合并AS大鼠模型的干预疗效,并从脂质代谢调节、AngII介导的氧化炎症反应系统、以及一氧化氮系统的改变等方面探讨原花青素在PCOS合并AS大鼠模型中防治AS的可能机制,为原花青素作为治疗PCOS合并心血管疾病的有效药物提供坚实的理论基础。

英文摘要: Polycystic ovary syndrome (PCOS) is one of most common disease in premenopausal women. Patients with early stage of PCOS are associated with atherosclerosis(AS), and the risk of cardiovascular disease is increased in the later period of PCOS. However , there are no any effective drugs for treatment of PCOS complicated with AS. Furthermore, the pathogenenis of angiotensin II (AngII) mediated oxidative stress and inflammatry response in PCOS with AS is not clear. Proanthocyanidins, an antioxidant which is a new, natural and effective antioxidant. In this study, we developed a PCOS complicated with AS animal model by high fat dieting for 16 weeks in DHT-induced PCOS rats.The radioimmuno assay, Western-Blot and Real time PCR were performed to look at the therapeutic effect of Proanthocyanidins to the PCOS with AS. We will further investigate the role of Proanthocyanidins in lipid metabolism, AngII mediated oxidative stress and Nitric oxide system. This will explore the underlying mechanisms of PCOS with AS, and provide the theoretic evidence for application of Proanthocyanidins in PCOS complicated with cardiovascular disease.
