中文摘要: 神经干细胞(NSC)在一定的条件下可以激活、增殖并分化为神经元、星形胶质细胞和少突胶质细胞,修复神经损伤,为治疗脑卒中开辟了新的途径。本研究基于神经血管单元(NVU)的网络靶点,采用体内试验(大鼠MCAO)和体外实验(低糖低氧损伤NSC)的方式,利用电镜、免疫组化、免疫荧光及PCR、Westernblot等技术,检测缺血侧海马神经元NGF、BDNF、GDNF、PDGF、VEGF、NF-200、GFAP、MBP、Nestin、PSA-NCAM、 NeuN的动态变化,同时检测JAK2、STAT3、STAT1、MKM4、MKK7、NFAT2、NFAT4、ATF2的表达水平,探讨清脑益元汤对内源神经干细胞增殖分化的诱导作用以及对外源性神经干细胞的归巢作用,探讨这些作用是否通过调控JNK或JAK-STAT信号转导通路来实现。旨在为清脑益元汤治疗缺血性脑损伤,重建神经功能提供实验依据。

英文摘要: Neural stem cells could be activated, proliferated and differentiated into neurons, astrocytes and oligodendrocytes in repairing nervous injury in certain conditions, which opens a new approach in the treatment of stroke. Based on the the view of target-network related to the NVU, this research choose the experiments which are both in vivo(MCAO in rats) and in vitro (low sugar, low oxygen for damaging NSC), use the technologies of Electron microscopy, immunohistochemistry, immunofluorescence, PCR, Westernblot and so on, in order to detect the dynamic changes of indicators in the ischemic side of hippocampal neurons as follows: NGF,BDNF, GDNF, PDGF, VEGF, NF-200, GFAP, MBP, Nestin, PSA-NCAM and NeuN; while the expressions of the following indicators are to be detected: JAK2, STAT3, STAT1, MKM4, MKK7, NFAT2, NFAT4, c-JUN and ATF2. It aims to explore the mechanisms on endogenous proliferation-differentiation and exogenous homing of neural stem cells with the Qingnao Yiyuan decotion, to explore whether these effects are passed by regulating JNK or JAK-STAT signal transduction pathway, and to provide a experimental proof for Qingnao Yiyuan decotion in curing ischemic brain injury and reconstructing nerve function.
