中文摘要: 早期诊断和有效治疗是癌症研究领域的热点和难点之一。金纳米探针既可以通过光声成像提高癌症诊断的灵敏度和准确度,又能利用光热效应和载药性能实现癌症的有效治疗。如何进一步构建多功能金纳米探针促进癌症的诊疗一体化应用,是目前研究的发展趋势。基于此,本项目将分别以金纳米棒和金纳米笼为载体,利用核酸的表面自组装修饰,通过药物分子的特异结合或内部装载,构建具有光声信号增强效果、高效光热转换能力、药物可控运输的多功能金纳米探针;通过搭建光声成像和双模式治疗相结合的技术平台,以肿瘤细胞、荷瘤鸡胚胎和荷瘤裸鼠三个不同层面为研究对象,系统评估多功能金纳米探针在光声成像引导下靶向治疗的性能,探讨诊断和治疗的关联机制和新方法。本研究有望为癌症诊疗一体化研究提供新技术手段,为推动纳米材料在临床中的应用发挥积极作用。

英文摘要: The early diagnosis and efficient therapy is one of the hot spots and difficulties in cancer research. Gold nanoprobes not only can improve cancer detection with high sensitive and specific by photoacoustic imaging, but also can realize efficient therapy through photothermal effect and drug delivery performance. The construction of multifunctional gold nanoprobes for cancer theranostics is the development trend of current research. Thus, this project will first synthesis gold nanorods and gold nanocages as nanocarriers, respectively. Then, nucleic acid will self-assemble on the surface of gold nanoparticles via Au-S bonding. Finally, anticancer drugs will be bound to the surfaces or encapsulated in the hollow interior of these nanomaterials to form multifunctional gold nanoprobes with photoacoustic enhancement effect, high photothermal conversion ability and controlled drug delivery. Photoacoustic imaging system based theranostic platform will be constructed to systematically assess the theranostic performance of these gold nanoprobes in cancer cells, tumor-bearing chicken embryo and mice model. The mechanism and method associated between diagnosis and treatment will be discussed. This project will provide a new technology in cancer theranostics, and will promote the development of nanomaterial in clinical applications.
