中文摘要: 大肠癌是我国发病率位居前三的恶性肿瘤,开发原位标记和检测分析大肠癌标志物分子的方法,及时、准确地判断其侵袭、转移能力,是大肠癌诊疗中亟待解决的关键问题。前期研究表明:整合素α3、α6 及β1表达水平与Dukes 分期、肿瘤分化程度、预后有关。因此,研究其在大肠癌中分布及表达量改变,对判断大肠癌的恶性程度、侵袭及转移能力具有重要的参考价值,是实现大肠癌早期诊断的有效途径。.本项目拟构建集荧光、CT及光声信号于一体靶向大肠癌整合素的Bi2S3/金属团簇复合纳米探针。依赖其荧光及金属丰度信号,在细胞水平实时、动态研究整合素表达量及分布的变化;利用一体化的荧光、CT、光声信号,在小动物活体水平研究不同恶性程度大肠癌组织中整合素分布及表达水平变化;分析其与大肠癌病理发展、转移相关性,为大肠癌早期诊断提供有效手段。在此基础上,利用探针光热转换、辐射敏感性实现大肠癌的光热、辐射协同治疗。

英文摘要: Colorectal cancer is among the top three of the incidence of malignant tumors, develop in situ labeling and detecting methods for colorectal cancer markers analysis, timely and accurately determine the invasion and metastasis ability of colorectal cancer, is urgently needed. Previous studies suggested: integrin α3, α6 and β1 expression levels correlated to Dukes stage, tumor differentiation, prognosis. Therefore, to study the distribution and expression of integrin in colorectal cancer, is important to determine the degree of malignancy, invasion and metastasis abilities of colorectal cancer. It is also an effective way to achieve early diagnosis of colorectal cancer..The project intends to design and screening of colorectal cancer-specific targeting integrin polypeptide sequence, and construct the multi-modal Bi2S3 / noble metal cluster nanoprobe. Based on the tunable fluorescence and the different metal content of the probe, we can dynamic study the distribution and the expression of integrin at the cellular level in real-time. Analyze the expression of integrin in different colorectal cancer subtypes associated with colorectal cancer development and metastasis; on the basis of the integrative fluorescence, CT, acoustics signal of the probe, through the multi-mode imaging modalities, observe the integrin distribution and expression levels in vivo, study the correlation between the pathological changes of colorectal cancer and the integrin expression status, and provide an effective protocols for the early detection and diagnosis of colorectal cancer. Furthermore, taking advantage of light-heat conversion property of the Bi2S3 the radiation-sensitive nature of the probe, we can achieve light and heat, radiation synergistic treatment of colorectal cancer.
