中文摘要: 抵抗细胞死亡是肿瘤的重要特性,也是肿瘤治疗失败的主要原因。ERBB受体酪氨酸激酶家族蛋白HER4对骨肉瘤可能具有重要的保护作用。本课题前期工作结果显示:HER4保护骨肉瘤在细胞应激状态下存活并抑制凋亡;HER4与调控细胞自噬水平有关;抑制HER4降低了骨肉瘤肺转移率,提示HER4对骨肉瘤细胞在不利环境下存活的重要性,然而其具体调控机理尚不明确。我们认为HER4可能通过调控自噬以保护骨肉瘤耐受细胞应激,从而逃避细胞凋亡。 基于以上工作基础,本课题拟通过一系列实验判断HER4在骨肉瘤中起保护作用的主导功能基团,并对其下游通路中可能与抑制凋亡相关的重要分子进行检测;对HER4调控自噬的通路和关键分子进行分析和验证;检测HER4对骨肉瘤肺转移和化疗敏感性的影响。本课题将解析HER4在骨肉瘤中维持肿瘤生存的作用机制,从而探讨HER4作为骨肉瘤治疗靶点的可能性。

英文摘要: Resisting cell death is one key hallmark of cancer. The ERBB receptor tyrosine kinase family member HER4 may play important pretoective role in osteosarcoma. Our previous work suggested that HER4 protects osteosarcoma cells from cellular stress and avoid apoptosis. HER4 may be related to the regulation of autophagy. Inhibition of HER4 decreased lung metastasis. We hypothesized that the function of HER4 to regulate cell autophagy is key to these findings. Based on the above observation, the purpose of this proposal is to determine which functional domain of HER4 is responsible for its protection effect in osteosarcoma. We want to analyze its potential downstream pathways and interacting molecules that relate with anti-apoptosis effect. Second, we plan to analyze how HER4 regulates autophagy, and through which key moleculars. Further, we will also measure the impact of HER4 on lung metastasis and chemosensitivity in osteosarcomas. Our study will clarify the role of HER4 in survival of osteosarcoma. Our findings will test the possibility for HER4 to become a new therapeutic target for osteosarcoma.
