中文摘要: 针对生物样品中痕量蛋白质鉴定困难的问题,本项目发展了一系列基于生物质谱的痕量蛋白质研究的新技术新方法。包括基于复合纳米材料富集低丰度蛋白质和质谱高灵敏鉴定的研究,基于复合纳米材料富集糖蛋白质/糖肽和质谱高灵敏鉴定的研究,基于复合纳米材料的低丰度蛋白质高效酶解研究和疾病相关的蛋白质组定量研究。通过本项目的研究,挖掘了一批有用的低丰度蛋白质和后修饰蛋白质,为重大疾病如大肠癌等的研究提供了翔实的数据。

英文摘要: The successful quanlification and quantification of low abundant proteins is difficult and of great importance in proteomics research. In this program, we have developed a series of mass spectrometry-based new methods and techniques for the research of low abundant proteins. We synthetized different composite nanomaterials for highly efficient enrichment of low abundant protein and glycoprotein/glycopeptide , and highly efficient proteolysis of low abundant protein. Disease-related proteome quantification were also included in this program. A lots of meaningful low abundant protein and post-translational protein were identified, which provided detailed data for research of disease such as colorectal cancer.
