中文摘要: 为进一步研究阿托伐他汀改善梗死微环境的机制,本项目基于我们课题组前期实验结果,假设阿托伐他汀通过促进心肌细胞自噬,抑制梗死微环境诱导的心肌细胞凋亡。拟通过体外细胞实验,培养乳鼠心肌细胞,通过缺氧无血清模型建立,模拟体内缺血再灌注损伤环境,通过观察他汀凋亡相关基因沉默心肌细胞及自噬相关基因沉默心肌细胞在缺氧无血清环境中的作用,研究他汀的抗凋亡机制,并进一步阐明相关信号通路。同时拟以动脉粥样硬化Wister大鼠100只为研究对象,建立急性心肌梗死模型,随机分为正常组、假手术组、急性心梗组、心梗+他汀组以及心梗+信号通路抑制剂组,于心梗造模术后1周及4周行心脏核磁共振检查,评价心功能;并于第4周磁共振检查后处死动物,取材左室心肌,行组织学和分子生物学检测,渴望证明上述假设,并阐明其机制。本研究原创性突出,设计新颖,具有重要的临床应用意义。

英文摘要: In order to investigate the mechanisms of atorvastatin improving post-infarcted environment, according to our previous results, we assume that atorvastatin could inhibit the apoptosis by activating the autophagy of cardiomyocyte. We establish the cardiomyocyte model of hypoxia and serum deprivation and use the siRNA transfection to make apoptosis or autophagy related gene silence to observe the statins effect on apoptosis and autophagy. Meanwhile, we will use the arthrosclerosis rats (n=100) for animal study and randomize into give groups: normal group, sham group, AMI group, AMI+atorvastatin group and AMI+compound C (the inhibitor of AMPK) group. All the animals will get MRI examination on the first and the forth week after AMI operation. After MRI test on the forth week post-AMI, animals will be sacrificed and token the left ventricular for histology and molecular-biology analysis in the purpose of testifying the hypothesis above. This original investigation is novelty designed and with great clinical significance.
