中文摘要: AGPase 是植物淀粉生物合成过程中的关键酶。本研究以公布的甘薯淀粉生物合成关键酶AGPase的两个小亚基和一个大亚基的基因序列为依据,设计了多对TRAP-PCR 引物,提取了36个淀粉含量特异的甘薯品种进行多态扩增,并研究了TRAP 标记多态性与淀粉率的相关性,筛选出适合的引物。在此基础上,提取了110个杂交后代DNA并进行多态扩增,测定了淀粉率,分析了两者间的相关性并建立了杂合体TRAP 多态性与高淀粉表型性状相关的生物信息学分析技术及其相应的数学分析模型。进而研究提出了包括亲本选配、后代实生苗苗期筛选的高淀粉育种新策略和新技术。本研究对于缩小甘薯育种田间试验规模,缩短育种年限,提高育种工作效率具有十分重要的理论和现实指导意义。

英文摘要: In this research, TRAP-RCR primers were designed according to the published sequence of two small subunits and a large subunit of AGPase, which is one of the key enzymes responsible for plant starch biosynthesis, TRAP-PCR amplification were performed on 36 parent sweetpotatoes with various starch content and then the primer relative to phenotypic characters of starch content was screened. Based on that, DNA were extracted from over 110 offsprings and TRAP-PCR was Performed, their starch content was also measured. By analying the results, a new bioinformatics analysis technique and their mathematical models to explain the relationships were established in this study. We also have developed new high starch breeding strategies and techniques for parent selection and early selection of seedlings. This study may have very important theoretical and practical significance for reducing the sweetpotato breeding scales of the field trials, shortening the breeding period and enhancing the breeding efficiency.
