中文摘要: 病毒来源的small RNA(vsiRNAs)在病毒与寄主的互作中具有重要的作用。目前,有关中国小麦花叶病毒(Chinese wheat mosica virus,CWMV)的分子生物学研究尚未深入开展,而对于该病毒来源的vsiRNAs及其与寄主的互作关系未见报道。在本项目的前期工作中,申请者构建了具有侵染活性的CWMV全长侵染性克隆,并利用small RNA测序、降解组测序、Northern-blotting的方法获得一个高丰度表达的vsiRNA-40及其可能寄主靶基因TaELP3。因此,本项目拟在前期工作基础上,利用5‘-RACE、瞬时表达技术、点突变技术以及反向遗传技术等各种分子生物学的方法系统的鉴定vsiRNA-40调控靶基因TaELP3对寄主抗病性的影响。本项目的开展有望为CWMV致病机理提供科学依据,从而为进一步分析vsiRNA与寄主的互作提供新的依据。

英文摘要: Virus-derived small interfering RNAs (vsiRNAs) at the core of plant-virus interaction. Now the pathogenic and molecular mechanisms of Chinese wheat mosica virus (CWMV) still remain little know. The interactions of the CWMV and its vsiRNAs has not been reported until now. Previously, the characterization of siRNAs derived from CWMV were analysised and the full cDNA clone was constructed sucessfully. Furthern research showed that the vsiRNA-40 target the TaELP3 gene by the degradome and small RNA sequencing. In this research we will used the Northern-blotting, 5'-RACE, transient expression technique and mutation technique to furthern indentify the vsiRNA-40 regulated the TaELP3.and its effect to host resistance. These results will contribute to a better understanding the CWMV pathogenic mechanism and provide a new sight to interaction of vsiRNAs and host.
