中文摘要: 近年来,高致病性流感频繁爆发、肆虐全球,严重威胁着人类的生命健康。现有医药技术在防治高致病性流感方面还十分薄弱,研制开发新型抗流感药物刻不容缓。我们在前期工作中对来源于石蒜的生物碱成分进行了初步生物活性研究,首次发现石蒜碱型化合物对多种流感病毒具有显著抑制作用。本项目在前期工作基础上,进一步对石蒜生物碱进行抗病毒活性评价,发现4种石蒜生物碱具有显著的抗流感病毒作用。作用机制研究显示,石蒜碱通过抑制流感病毒RNP复合物的出核而发挥抗病毒作用。本研究发现石蒜碱是具有特殊作用机制的抗流感病毒小分子物质,为寻找和发现新型高效抗流感药物提供了新思路。

英文摘要: The frequently outbreaks of highly pathogenic influenza become a serious threat to human health and life in recent years. Research and development of new anti-influenza drugs become a very urgent task due to the lack of efficacy of the existing medicines. In our previous study, the biological activities of lycorine-type alkaloids were evaluated, led to the findings that these alkaloids displayed moderate to potent antiviral activities against IFVs. These observations indicated lycorine-type alkaloids could be potential anti-IFV agents. Based on the previous results, the inhibitory activities on IFV of lycoris alkaloids were further investigate. Four alkaloids displayed potent antiviral activities. In addition, mechanism investigation demonstrated that lycorine could inhibit the RNP complex entering into the cytoplasm during a single round of replication. These results will be benefit for the development of new anti-IFV drugs.
