中文摘要: 本项目旨在分子层面上研究高分子电解质聚合物(polyelectrolyte)透明质酸在多价离子溶液界面上的拓扑构象及分子间作用力的变化。将透明质酸通过物理吸附、化学修饰高分子表面及基层表面化学锚定等三种方法固定在界面,首先利用原子力显微镜(AFM),透射电镜(TEM),椭圆偏振仪,动态光散射等手段检测透明质酸层的表面性质,吸附行为和分子折叠的二级结构,明确透明质酸层的表面电荷与拓扑结构之间的关系。然后通过测量高分子层之间的表面力与反应自由能,揭示溶液环境和抗衡离子,尤其是二价离子对透明质酸分子层形态和功能的影响。最后总结分析,建立分子模型,阐明多价离子对高分子的构效影响以及对高分子层之间的表面力和反应自由能的调控机理,明确抗衡离子的类型与分子表面电荷及界面的拓扑结构之间的关系,拓宽和完善DLVO理论在高分子电解质领域的应用,为未来新型高分子材料的设计奠定基础。

英文摘要: The objective of this project is to study the topological behavior and intermolecular forces of polyelectrolyte Hyaluronic acid in multivalent counterion solution at surfaces. Hyaluronic acid would be fixed on the solid/liquid interface through three ways - physical adsorption, chemical modification of the polymer surface and polymer's chemical anchoring to the substrate. The polymer-coated surfaces can be characterized through atomic force microscopy (AFM), transmission electron microscopy (TEM), ellipsometry, dynamic light scattering (DLS) in order to determine the surface properties, adsorption activities and secondary folding of Hyaluronic acid and illustrate the relationship between polyelectrolyte surface charges and its topology prepared as above. Next, the surface forces and interaction free energy between Hyaluronic acid layers would be measured to investigate the mechanism of how the solution conditions and counterions, particularly divalent ions, affect the conformation and interactions of Hyaluronic acid layers. By establishing a model system and identifying the relationship between multivalent ions and polyelectrolyte interfacial topology, the understanding and application of charged polymer can be expanded further than the current DLVO theory.
