中文摘要: 骨关节炎是一种常见的关节退行性疾病,已成为影响患者生活质量的一个严重问题,其最显著临床特征即为疼痛。增强痛觉神经元兴奋性是痛觉形成的分子基础,而离子通道的功能活动是神经元产生兴奋性的分子基础。在前期工作发现Kv7/M通道开放剂Retigabine能够有效缓解骨关节炎疼痛基础之上,本申请拟深入研究骨关节炎模型背根神经元Kv7/M通道表达与功能,背根神经元的兴奋性及其与Kv7/M通道电流的关系,最后探讨Kv7/M通道功能调节对大鼠骨关节炎疼痛行为的影响。此研究对系统深入认识Kv7/M通道在骨关节炎疼痛中的作用及机制具有重要意义,同时为临床治疗骨关节炎疼痛提供新的靶点和新的科学思路。

英文摘要: Osteoarthritis is a common degenerative joint disease, which has become a serious problem affecting the quality of life of patients. The most notable clinical feature of osteoarthritis is the pain. The increase of the excitability of nociceptive neuron is fundamental mechanism for pain, and the activity of ion channels is the molecular basis for the excitability of neurons. Our previous work showed that Kv7/M channel opener retigabine significantly relieved the osteoarthritis pain. In this work,we will further investigate the expression and function of Kv7/M channel in DRG neuron of osteoarthritis model, the excitability of DRG neurons and its relationship with Kv7/M channel, and the effect of Kv7/M channels modulation on rat osteoarthritis pain. This study will show important meaning on Kv7/M channels involved in osteoarthritis pain and give light to new targets and new ideas for clinical treatment of osteoarthritis pain.
