中文摘要: 本项目针对组织工程血管、人工血管移植后发生再狭窄难题以及锌指蛋白A20在抑制动脉粥样硬化中的重要作用。以锌指蛋白A20为靶基因,计算机分析其序列,设计合成能与A20基因启动子区18bp序列特异结合并能高效启动内源性A20基因表达的人工锌指转录因子,再在其N-末端加上9个精氨酸穿膜序列,得到人工锌指转录因子ZF-AFT,观察ZF-ATF活性蛋白对内皮祖细胞、平滑肌细胞的穿膜效果及功能。将人工转录因子ZF-ATF PEG化,与光交联壳聚糖纳米材料复合。在构建的血管支架中,用PEG化ZF-ATF蛋白等复合光交联纳米材料对血管腔进行修饰,在自行构建的血管生物反应器中构建组织工程血管,动物实验验证其抗动脉粥样硬化效果。本课题的完成将检测PEG化人工转录因子与光交联纳米材料在血管表面修饰中的作用,为人工血管、组织工程血管、血管内支架抗动脉粥样硬化提供具有知识产权的新方法。

英文摘要: To avoid atherosclerosis may be to conduct local treatment with tissue engineering blood vessels carrying functional genes. A20 gene is a functional gene that is useful for suppressing atherogenesis and graft atherogenesis. In order to create A20 gene site-specific zinc finger DNA-binding protein, the sequence of A20 gene promoter was analyzed with bioinformatics means, designed the amino acid sequence of zinc finger protein predicted to bind to 18 bp sequence of the A20 gene promotor. Thus, artificial zinc finger transcript factor were constructed by adding 9 Arginine to N-terminal end of transcript factor. To observe the function of ZF-ATF for protecting endothelial progenitor cells and smooth muscle cells. To add PEG to ZF-ATF and combinate the photocrosslinking chitosan. We constructed TEBVs using PEG-ZF-ATF regulation in a new bioreaction. The effect of anti-artherosclerosis of TEBV was checked by animal model. The results will check the function of PEG-ZF-ATF artifical blood vessel, it will provide a new anti-artherosclerosis method for artifical blood vesel,TEBVs and endovascula stent.
