中文摘要: 线粒体被称为细胞能量工厂,是细胞凋亡内途径的调控中心。在脊椎动物中线粒体在凋亡信号的刺激下,跨膜电位下降,外膜通透化,释放膜间隙蛋白如细胞色素c等凋亡因子到细胞质中,它们或激活胱天蛋白酶或独立破坏核染色质,导致细胞凋亡。在昆虫中该途径仍然存在着许多争议。本课题组前期研究发现喜树碱诱导的甜菜夜蛾细胞凋亡为线粒体调控的内途径。因此,本项目围绕线粒体,采用细胞生物学、分子生物学、免疫学和荧光标记等技术研究喜树碱诱导的细胞凋亡过程中线粒体形态、结构和功能的变化;线粒体结构和功能改变在细胞凋亡过程中的作用;以及凋亡过程中线粒体结构和功能改变的机制。从而阐明喜树碱诱导的昆虫细胞凋亡过程中的线粒体的作用和机理。这对于研究线粒体的昆虫毒理学意义和喜树碱的杀虫活性机制具有重要的价值,同时该研究项目将提高我国植物源活性物质的研发水平,而且对于发现生态安全性特异靶标和环境和谐性农药都具重要的探索性。

英文摘要: Mitochondria,the cellular power plants , is the key mediator of apoptosis. In the intrinsic pathway of apoptosis, mitochondria sense catastrophic cellular changes and irreversibly commit cells to apoptosis by releasing death factors such as cytochrome-c into cytosol. The release of these proteins is a consequence of the integrity of the mitochondrial outer membrane permeabilization (MOMP). So far, this process has only been validated in vertebrates; it is debated in insects. Our previous studies showed that mitochondrial involved in camptothecin-induced apoptosis in Spodoptera exigua IOZCAS-Spex-II cells. Therefore, we will study mitochondrial structure and function changes with the empirical methods of immunology, molecular biology and biochemistry after Spodoptera exigua IOZCAS-Spex-II cells treated by camptothecin in this project; investigate the effect and mechanism of mitochondria changes during the apoptosis. Results of these will map out the so called intrinsic pathway of apoptosis regulated by mitochondrial in insect cell apoptosis throughout the process from initiation of the first apoptotic signal to the final consequential breakdown of cellular integrity. These studies will provide the new insights to explore environment-friendly pesticides and are important for researches focused on molecular mechanisms of insect cell apoptosis and insect toxicology.
